Abstract

The preclinical drug development is based on drug metabolism and pharmacokinetics (DMPK), which defines the efficacy, safety, and dosage profile of therapeutic agents. Analytical chemistry has been crucial in the understanding of absorption, distribution, metabolism, and excretion (ADME) in animal model studies and therefore offers a useful way of understanding pharmacokinetic pathways and metabolic mechanisms. This review examines the influential works of analytical chemistry in the field of DMPK research with focus on chromatographic and spectroscopic methods, bioanalytical methods, and more advanced metabolite profiling using the advanced mass spectrometry. It also considers the weaknesses of current methods of analysis, issues in animal model research, and how current methods can be improved to achieve more accurate, sensitive, and translational quality of analytical analysis in preclinical drug metabolism and pharmacokinetics.
